SCOGFP, A SYNTHETIC GREEN FLUORESCENT PROTEIN GENE OPTIMIZED FOR EXPRESSION POTENTIAL IN SCHIZOPHYLLUM COMMUNE

Green Fluorescent Protein (GFP) expression has been previously reported in the basidiomycete Schizophyllum commune.  However, taking advantage of fluorescent protein technology for gene expression, protein localization and protein-protein interaction studies has proven elusive in our hands.  In an attempt to overcome this problem we have designed and created ScoGFP, a S. commune codon-optimized version of Emerald GFP.  In addition to the optimized coding sequence, the synthesized gene contains sixty bp of upstream sequence and two introns derived from the sequence of the highly expressed Translation Elongation Factor 1a gene of S. commune.  Current progress in analyzing the expression potential of this gene in Schizophyllum commune transformants will be presented.
